A profile grants object and field-level permissions plus system permissions — create/read/edit/delete rights, page layouts, apps and tabs — and every user must have exactly one. A role sits in the role hierarchy and governs record-level visibility, letting a user see the records owned by people below them; roles are optional and are used with sharing rules rather than permissions.
